Inhibitory effect of Butyl alcohol extract of BaiTouWeng decoction on Candida albicans cell wall

Abstract:

Objective To investigate the inhibitory effect of Butyl alcohol extract of BaiTouWeng decoction (BAEB) on Candida albicans cell wall. Methods The effect of BAEB on Candida albicans cell activity was detected by spot assay. The changes of β-1,3-glucan and chitin was measured by flow cytometry and microplate reader after BAEB treatment. QRT-PCR was used to detect the expression of β-1,3-glucan related genes FKS1 and chitin related genes CHS1, CHS2, CHS3 and CHS8 in Candida albicans cell wall. Transmission Electron Microscopic was used to observation the effect of BAEB on cell wall structure of Candida albicans. Results The Candida albicans activity decreased with the BAEB intervention. Exposure of chitin and β-1,3-glucan increased gradually in the 256, 512 and 1 024 μg/mL BAEB groups (P<0.05). The levels of FKS1, CHS1, CHS2, CHS3 and CHS8 were down-regulated by 5.57, 1.35, 3.29, 4.47 and 3.00 times in the group of 1 024 μg/mL BAEB treatment respectively. TEM showed the cell wall structure of Candida albicans was damaged. Conclusions BAEB can inhibit the expression of β-1,3-glucan, chitin and their biosynthesis related genes through the increase of β-1,3 -glucan and chitin exposure, So as to disrupt Candida albicans cell wall integrity.
