肺泡上皮细胞应对烟曲霉感染过程中胞内双特异性磷酸酶表达规律的研究

Study on the expression of dual specificity phosphatase in alveolar epithelial cells during Aspergillus fumigatus infection

摘要:

目的 研究肺泡上皮细胞应对烟曲霉感染过程中双特异性磷酸酶(dual specificity phosphatases, DUSPs)的表达规律和炎症因子释放规律。方法 在烟曲霉感染肺泡上皮细胞过程中,应用ELISA方法检测细胞上清中炎症因子的释放情况;应用Western blot检测胞内DUSP2、DUSP6和DUSP8的表达规律以及p65、ERK1/2和p38的表达规律和磷酸化水平。结果 烟曲霉B5233孢子感染肺泡上皮细胞过程中,细胞上清中炎症因子TNF-α、MCP-1、IL-12p40、IL-1β、IL-6、IL-8的含量均逐渐显著升高;胞内DUSP2的表达逐渐显著升高,DUSP6的表达逐渐显著下降,而DUSP8的表达无明显变化;ERK1/2和p65磷酸化水平显著升高。结论 肺泡上皮细胞应对烟曲霉感染过程中显著上调胞内DUSP2的表达,抑制DUSP6的表达,而对DUSP8的表达无显著影响,同时胞内NF-κB信号和MAPK信号均被激活,释放多种炎症因子。

Abstract:

Objective To study the expression of dual specificity phosphatases (DUSPs) in alveolar epithelial cells in response to Aspergillus fumigatus infection and the release of inflammatory factors. Methods During Aspergillus fumigatus infection, ELISA method was used to detect the release of inflammatory factors in the supernatant of alveolar epithelial cells, western blot was used to detect the expression of DUSP2, DUSP6 and DUSP8 and the expression and phosphorylation level of p65, ERK1/2 and p38 in alveolar epithelial cells. Results During the infection of alveolar epithelial cells by Aspergillus fumigatus B5233 conidia, the content of TNF-α, MCP-1, IL-12p40, IL-1β, IL-6 and IL-8 in the cell supernatant gradually increased significantly. And the expression of intracellular DUSP2 gradually increased, the expression of DUSP6 gradually decreased, but the expression of DUSP8 did not change significantly. The phosphorylation level of ERK1/2 and p65 increased significantly. Conclusion In response to Aspergillus fumigatus infection, the expression of DUSP2 increased and the expression of DUSP6 was inhibited in alveolar epithelial cells, but there was no change about DUSP8 expression. At the same time, intracellular NF-κB signal and MAPK signal were activated to release a variety of inflammatory factors.
