1例阴道定植阿萨希毛孢子菌的基因鉴定及随机扩增多态性DNA分析

Vaginal colonization of Trichosporon asahii in an asymptomatic woman using gene identification and random amplified polymorphic DNA

摘要:

报道1例阿萨希毛孢子菌阴道定植病例。1例25岁健康妇女偶有阴道分泌物稍增多及轻微瘙痒。妇科检查见少量淡黄色均质化分泌物附着于阴道黏膜,阴道分泌物Nugent评分3分。对两次阴道分离菌株进行形态学观察、温度试验、尿素酶试验、药敏试验、3个rDNA基因测序及随机扩增多态性DNA(RAPD)分析。间隔3个月两次阴道分泌物培养均分离出土黄色、表面皱褶的酵母样菌落。该菌在25~37℃生长良好,最佳生长温度为27℃。小培养可见分隔菌丝、假菌丝、关节菌丝、分生孢子、关节孢子和芽生孢子。尿素酶试验阳性。3个rDNA基因测序和RAPD分析显示两次分离菌株为同一种阿萨希毛孢子菌。E-test和Neo-Sensitabs药敏试验发现本菌株对伏立康唑敏感。随访3个月患者仍无明显症状。

Abstract:

To report a case of vaginal colonization of Trichosporon asahii.A 25-year-old healthy woman occasionally developed mild itching and vaginal discharge.Gynecological examination showed a small amount of yellowish homogenous vaginal discharge.Nugent score of vaginal discharge was 3.Two strains isolated from vaginal discharge were analyzed by phenotypic observation,temperature test,urea enzyme test,antifungal susceptibility test,3 rDNA gene sequencing and random amplified polymorphic DNA (RAPD).The earthy yellow and wrinkled yeast-like colonies were twice isolated from the vaginal discharge at 3-month intervals.The isolate grew well at 25-37℃,with the optimal growth temperature of 27℃.Slide culture revealed septate hyphae,pseudohyphae,arthrofilaments,conidia,arthroconidia,and blastospores.The strain was urease positive.Both isolates belonged to the same strain of T.asahii using 3 rDNA gene sequencing and RAPD analysis.E-test and Neo-Sensitabs results revealed that the isolate was sensitive to voriconazole.The woman remained asymptomatic during 3-month follow-up.
