天然产物TS抗念珠菌作用研究

In vitro activity of triterpene saponins from tea plant (Theaceae) against Candida species

摘要:

目的 探讨天然产物TS体外抗念珠菌作用。方法 采用微量液基稀释法测定最低抑菌浓度(MIC),琼脂稀释法测定最低杀菌浓度(MFC),透射电镜观察TS对念珠菌超微结构的影响。结果 TS抗白念珠菌的MIC范围0.08~0.32 mg/mL,抗光滑念珠菌MIC范围0.08~0.16 mg/mL,抗热带念珠菌MIC范围0.32~0.64 mg/mL,抗近平滑念珠菌MIC 0.32 mg/mL,抗克柔念珠菌MIC 0.32 mg/mL。对白念珠菌具有杀菌作用,MFC 0.64 mg/mL。透射电镜下可见:细胞壁皱缩、破裂;细胞核异常;细胞质中可见较多空泡状结构;细胞中无完整细胞器结构。结论 TS具有明确的抗念珠菌作用,可使其形态和超微结构发生明显的改变。

Abstract:

Objective To explore the antifungtal activity of TS from Camellia sinensis L.against Candida.Methods The minimal inhibitory concentration (MIC) was tested by broth microdilution method based on CLSI (Clinical and Laboratory Standards Institute).The minimal fungicidal concentration (MFC) was tested by agar plate dilution assay.The ultrastructure of Candida albicans treated with TS was observed by transmission electron microscopy.Result The range of inhibitory concentration of the antifungal activity against Candida albicans were from 0.08 mg/mL to 0.32 mg/mL.The minimal fungicidal concentration against Candida albicans was 0.64 mg/mL.Candida albicans treated with TS showed that the cell wall shrinked and ruptured,the cell nucleus had the abnormal morphology and the cell organelles were imcomplete as well as vacuoles were formed under TEM.Conclusion TS has antifungal activity against Candida species in vitro.
